Tuesday 7th May
The day will consist of morning and afternoon session, each three hours long, with each of the sessions focusing on three staining techniques regularly required to be submitted by UK NEQAS CPT participants for EQA assessment.
The day is designed to be relaxed, informal experience, in which interaction and questions will be actively encouraged to hopefully stimulate conversations that benefit the speaker and the participants.
Each of the six staining techniques will have one hour dedicated to it in which the speaker will explore:
(a) the chemistry and mode of action of a technique
(b) optimal staining and expression
(c) guidance on troubleshooting and how to avoid/resolve staining issues
Morning Session (09:30 -12:30)
H&E, Reticulin, and EVG/HVG staining techniques.
Break (12:30 -13:00)
Afternoon Session (13:00-16:00)
AB/PAS (DPAS), Gram, and Copper Associated Protein staining techniques.
At the end of each session there will hopefully be some time for additional questions from participants
The speaker’s presentations will be a mix of text, diagrams, and images of staining techniques.
A few days after the event, all participants will be sent pdf versions of all the presentations they attended
Participants can make their own additional notes during sessions without the concern that that they must copy down what they see on screen!
Wednesday, 8th May
The day will consist of morning and afternoon session, each three hours long. The morning session will explore technical considerations that affect ICC staining quality, and the afternoon session will be an entirely image based discussion. The day is designed to be relaxed, informal experience, in which interaction and questions will be actively encouraged to hopefully stimulate conversations that benefit the speaker and the participants.
Morning Session (09:30-12:30)
During this session the speaker will explore the range of factors that can influence the quality of ICC staining, provide some guidance on troubleshooting and how to avoid/resolve staining issues. Topics covered will include:
Cold ischaemia
Fixation
Section quality
Epitope stability
Optimal epitope retrieval
Antibody optimisation
Selection and use of control tissue
Control storage
IQA, EQA
Automated instrument issues
Staining artefacts.
Break (12:30-13:00)
Afternoon Session (13:00-16:00)
During this session participants will take part in a screen based session with the speaker, systematically assessing and discussing images of ICC stained slides generated from a wide range of clinically relevant antibodies. For each of the antibodies the expected staining expression patterns, expected cellular locations and their clinical diagnostic uses will be discussed.
As well as routine clinical diagnostic antibodies, the session will also discuss stained images from drug treatment associated antibodies, including HER2, PD-L1, MMR and ER. Discussions will look at the interpretation of these antibodies, their problems, and the use of assay controls.
The final part of the session will discuss poor ICC staining and staining artefacts.
At the end of each session there will hopefully be some time for additional questions from participants.
